'Maiden' Inca Mummy Suffered Lung Infection Before Sacrifice

The Maiden mummy of a 15-year-old girl who was sacrificed some 500 years ago suggests she likely suffered from a lung infection at the time of her death, scientists reported July 25, 2012.

The so-called Maiden mummy of a 15-year-old Incan girl who was sacrificed 500 years ago is giving up some secrets, revealing the teenager suffered from a bacterial lung infection at the time of her death, scientists report Wednesday (July 25).
The researchers analyzed tissue proteins, rather than DNA, from the Maiden and another young Inca mummy who died at the same time.
Over the last decade, DNA techniques have proven useful in helping solve ancient mysteries, such as how King Tut died. But these techniques aren't without faults. For example, finding evidence of a malaria-causing parasite in King Tut's system doesn't necessarily mean the Egyptian king suffered any malaria symptoms. Additionally, the environment can easily contaminate DNA samples, if researchers aren't careful.

On the other hand, analyzing a sample's proteins, which are less susceptible to environmental contamination, yields a whole different set of information. "Being the expression of DNA, proteins really show you what the body is producing at the time when the individual is being sampled — or, in our case, at the time of death," study researcher Angelique Corthals, a forensic anthropologist at the City University of New York, told LiveScience. In particular, proteins can tell you if the body's immune system has activated to fight a disease, she added. [The Mummy Quiz: Test Your Smarts]

In their study, Corthals and her colleagues took lip swabs from two Andean Inca mummies, a 7-year-old boy and "the Maiden," as well as samples from the boy's bloodied cloak. The two child mummies, discovered in 1999, were originally buried on the summit of the Argentinian volcano Llullaillaco, 22,100 feet (6,739 meters) above sea level, after being sacrificed in a ceremonial ritual.

Past research found the boy and girl had been fattened up before sacrifice, being fed a typical peasant diet of potatoes and other common vegetables up until a year before their sacrifice, when evidence suggests they were given "elite" foods like maize and dried llama meat.

"What I really wanted to do originally was see where the blood I found on the mummies' clothing and lips came from," Corthals said. "But we found a whole lot more than we were expecting."
Archaeologists also found a third mummy, a 6-year-old girl, along with the other two. This mummy appears to have been struck by lightning, which could potentially interfere with test results, so Corthals and her team didn't take any samples from it.

Lung infection found
The researchers used a technique called shotgun proteomics. They placed their samples into a device called a mass spectrometer, which broke all of the sample's proteins into their constituent parts, amino-acid chains. Sophisticated software compared these parts with existing proteins of the human genome to determine the actual proteins in the samples, Corthals explained. "You couldn't use this technique for an organism that we don't have the complete genome for," she said.
They found that the Maiden's profile of proteins match that of a chronic respiratory infection patient. X-rays taken of the Maiden's lungs after she was discovered also showed signs of a lung infection. To see if the Maiden was harboring anything that could cause such an infection, they turned to DNA analysis and discovered evidence of bacteria in the genus Mycobacterium, which is known to cause upper respiratory-tract infections and tuberculosis (TB). Statistical models suggested the bacterium falls into the cluster group that causes TB, but the exact species isn't known, likely because its DNA hasn't been sequenced yet.
The Llullaillaco boy didn't have signs of disease or pathogenic bacteria.
The research shows that shotgun proteomics can play a critical role in determining the disease or death in archeological, medical and criminal cases, Corthals said, adding that the method may even be able to determine which pathogen is the killer in a case of multiple infections. For now, Corthals is interested in seeing whether the technique can be used with less pristine samples, such as skeletal material or Egyptian mummies.
Down the line, the protein technique's utility will likely go beyond just archaeology, researchers said. "I expect [the method's] biggest impact will be in criminal forensic science," Corthals said.
The new study is detailed online today (July 25) in the journal PLoS One.

Your feet are at a high risk of contracting diseases in the monsoon season. Dr. Govind Bisht, Senior Consultant, Podiatry with Delhi based Sri Balaji Action Medical Institute shares daily footcare tips to prevent diseases and build up of bacteria.

Apply moisturiser:

Dr. Govind Bisht gives you an easy tip to fight fungal infection, "Before going to sleep during monsoon, apply moisturiser containing 7% to 10% of urea and paraffin on your feet. This will help you in getting rid of the dead skin (keratolytic effect) while keeping the feet clean, smooth, and allergy-free. Avoid putting moisturiser between the toes as web spaces have moist skin and putting moisturiser will increase dampness and may invite fungal infection."

Trim your nails:

This is one basic personal hygiene tips everyone should follow as it will prevent build up of germs, Dr. Bisht explains, "Long nails pull and store ample of dirt and bacteria. Always trim your nails in straight line and file the corners. Avoid cutting too deep and leaving corners with sharp margins may lead ingrown toes nails, well kept nails will help in keeping away germs from the feet."

Get a pedicure done:

Mani-pedi may be a fad among women, but a pedicure can help you prevent accumulation of bacteria. Dr. Bisht says, "Take out some time from your busy schedule and visit the nearest parlour for a pedicure, specifically during rains. Make sure the instruments and the place is neat and clean. Avoid pedicure from unhygienic places, it may be source of fungal and bacterial infection."

Home remedy for foot care:
He gives you a home remedy for pedicure, "If your budget does not permit, do it the easy way at home. Dip your feet in lukewarm water for 15 minutes and dry it with a soft cotton towel. Do not forget to add 2-3 drops of antiseptic liquid in the water. Diabetic patients should avoid putting feet in warm water, at times because of loss of sensation they may land in burn injuries."

Clean and keep your feet dry:

Monsoon season propels the growth of bacteria and germs, therefore Dr. Bisht adds that it is important to keep your feet dry, "It is a simple yet an essential thing to wash your feet after an outing during monsoon. As the season gives rise to fungus, bacteria, and germs, it becomes a necessity to clean your feet immediately after returning from office or school. Following the clean-up, pat them dry. Wear shoes rather than sandals while going out as shoes give the full protection compared to the sandals which make the feet wet, resulting in fungal and bacterial infections."

Foot Scrubbing:

Wash out all dead cells to prevent any fungal infection, Dr. Bisht, Podiatrist explains, "When you go for bathing, soak your feet in the water about 5 to 10 minutes. Add any soap liquids, shampoo to the water. After 10 minutes wash your feet with the scrubbers made for feet to chuck out the dead or damaged skin."

Note: Any sign of fungal or bacterial infection as redness, swelling, itching, and warm skin associated with pain without injury must not be neglected and a visit to physician is advisable for proper diagnosis and treatment.

It is always best to consult your physician or orthopaedist for a list of low-impact exercises suitable for your age as well as the types of pain that you have.

Stretch: Apart from exercise, stretching is vital for preventing back pain and staying flexible. Stretching is helpful in recovering from a back injury. Always stretch before exercise or other strenuous physical activity and also before bed.

Sleep well: Always sleep on a firm surface for back pain relief rather than soft mattresses that push your back out of alignment. Also, sleep on your side to shrink any curve in your spine.

Maintain good posture: Poor posture can lead to back pain or other health issues. If you are standing, stand up straight and keep your weight balanced on your feet. Make sure that your feet are shoulder-width apart and your knees slightly relaxed. Your shoulders should be straight and are in a straight line with your ears and hips.

While sitting, sit in a chair that is designed to keep your back straight. Switch your sitting positions often and taking periodical walk around the office will help relieve stress. If you need to sit for a long period of time, rest your feet on a low stool or on a foot rest.

Keep a healthy weight: Eat a healthy diet filled with fruits and vegetables and low on processed foods to avoid excessive weight. Staying fit is one of the best remedies for back pain as extra pounds can stress your back causing further hurt.

Quit smoking: Smoking doesn’t just damage your lungs, it can also impair your back.
Smoking cuts blood flow to the lower spine and causes the spinal discs to degenerate.
Studies have shown that smokers are more likely to have back pain than their peers who do not smoke.
